How do transport vesicles select their contents, bud, translocate through the cytoplasm, and then fuse with their targets? We study the Ras-like Rab GTPases--how they serve as master regulators of all receptor trafficking events. We are using automated microscopy, live-cell fluorescent biosensors and perturbations of predicted signaling proteins to systematically dissect signaling networks. Thomas Südhof is interested in how synapses are formed, how presynaptic terminals release neurotransmitters at synapses, and how synapses become dysfunctional in diseases such as autism or Alzheimer's disease.
